一、課程說明(Course Description):
The Yeast Genetics and Genomics course is designed to teach students various approaches needed to dissect complex problems in budding yeast Saccharomyces cerevisiae. Classical genetic approaches, such as the isolation and characterization of mutants, complementation, tetrad analysis, and mitotic recombination will be emphasized. Molecular genetic techniques, including yeast transformation, gene replacement with plasmids and PCR, construction and analysis of gene fusions, and generation of mutations in cloned genes, will also be addressed. Furthermore, modern cytological approaches such as epitope tagging and imaging yeast cells using indirect immunofluorescence, GFP-protein fusions and a variety of fluorescent indicators for various subcellular organelles will be covered in this course. These classical, molecular and cytological yeast approaches will benefit students to gain insights in genetic suppression and synthetic lethality in order to identify and interpret various kinds of genetic interactions. Latest papers on fundamental aspects of yeast genetics will also be discussed along with the corresponding lectures.
